Research in the Gayther laboratory is focused largely on understanding the underlying causes of ovarian cancer initiation and development. The overall approach of this research program is to integrate genomics and epigenomics analyses to identify molecular markers associated with disease, with cell biology modeling studies to validate the role of novel molecular markers in disease biology. Working under direct supervision of the Project Lead and the Center Director, the Research Associate I (RAI) will perform a variety of routine experimental protocols and procedures to support the objectives of one or more laboratory research projects in a specific area of research. The research projects are focused on developing induced pluoripotent stem cell (iPSC) lines from different normal human tissues and then differentiating them into different prhnotypes, including breast, ovarian, prostate, pancreatic and bladder cancer precursor tissues. Differentiated models will then be phenotpyped and molecularly profiled using single-cell methodologies. The RAI will assist in these research activities as well as in general laboratory activities, including maintenance of reagents and stocking of supplies; maintaining, updating, and keeping accurate and detailed records of experiments and results; assisting in the operation of specialized equipment and machinery; observing safety standards and procedures; performing routine micro- and molecular biology procedures including, but not limited to processing of blood and other biospecimens into different components, protein extraction and Western Blot analysis, immunohistochemistry, DNA/RNA extraction, PCR, cDNA preparation, cell culture, cloning of genetic fragments in bacteria. This position does not have supervisory responsibilities.
Primary Job Duties and Responsibilities:
- Performs a variety of routine laboratory tasks and procedures.
- Keeps accurate and detailed project records of experiments and results.
- May assist with animal husbandry.
- Maintains lab equipment and related records.
- Transports, processes and logs samples.
- Maintains computer database with relevant clinical information.
- Performs lab maintenance duties, including glassware cleaning and sterilization.
- Maintains stocks of general lab materials, and places orders for lab equipment and supplies.
- Assists new students and fellows at the laboratory.
- Assists in the operation of specialized equipment and machinery.
- Observes and complies with safety standards and procedures.
Department-Specific Responsibilities:
- Safely handle and process fresh human biospecimens.
- Safely and manually transport human biospecimens between hospital clinics and laboratories.
- Timely processing of samples strictly according to protocol (e.g. centrifugation-based processing of blood samples into buffy coat and plasma and aliquoting).
- Processing of formalin-fixed paraffin-embedded (FFPE) tissue samples.
- Extraction and quality control checks of DNA, RNA, circulating tumor DNAs, etc from human biospecimens.
- Various polymerase chain reaction (PCR)-based applications, Real-time PCR, and Sterile cell culture techniques.
- Rigorous upkeep of research databases, sample biobanking, and laboratory notes.
- Stock-taking and ordering laboratory supplies, Assembling sample collection kits.
- Safe handling of shipments on dry ice and Escherica coli bacteria for cloning DNA fragments.
Education:

Bachelors Degree in a science related field required.
Experience and Skills:
- No experience required. One or more years of practical experience working in a molecular biology/pathology/microbiology/genetics laboratory is preferred.
- Understanding of general research objectives.
- Working knowledge of database management, personal computers, and presentation software.
- Familiar with routine laboratory procedures and experimental protocols.
- Must possess computer skills to include, but not limited to, Excel and Word.
